Generation of viral particles
For biological safety all viral particles produced in the Virus Transduction Service are replication incompetent to prevent the formation of an infectious wildtype virus. For packaging of the expression constructs we use a 2nd-generation system consisting of 3 plasmids. The functional viral components are subdivided onto different plasmids:
- Lenti-or retroviral transfer plasmid
- Packaging plasmid (e.g Gag, Pol, Rev, Tat)
- Envelope plasmid (e.g pVSV-G)

The Virus Transduction Service currently does not intend to produce large amounts of high titer virus stocks or store larger amounts of viral supernatant. Therefore we have to dispose the viral particles 4 weeks after delivery of the transduced cell line.
