Tight regulation of the spatial and temporal organization of chromatin topology is essential for normal development and a driver of cancer when dysregulated. Epigenetic modifications, including histone modifications, DNA methylation and nucleosome positioning, are essential in orchestrating the temporal regulation of gene expression mediated by the 3-dimensional chromatin conformation. In cancer, single nucleotide mutations and large structural variations affect epigenetic regulation, leading to changes in gene expression networks and altered signaling pathways that drive or promote tumorigenesis. In recent decades, (epi)genetic alterations involving cis-regulatory elements have been recognized as major players in tumorigenesis. This symposium will provide an up-to-date overview of the importance of chromatin topology and regulatory element alterations in cancer and discuss how this information can be relevant for cancer therapy.
