Virus Transduction Service

Applications

The goal of the Central Virus Transduction Service is to provide fast, cost-effective and uncomplicated access to virus transduced cell lines. The Virus Transduction Service offers the packaging and transduction of user-supplied lenti- or retroviral constructs and cell lines as well as the complete S2 project documentation.Custom generation of expression constructs in our predefined set of fully sequenced lenti- or retroviral expression vectors are available in combination with the Gateway-Shuttle Service. The Facility authorized and fully equipped for production of small scale lenti- or retroviral particles. The Virus Transduction Service does not intend to produce large amounts of high titer virus stocks or store large amounts of viral supernatant.

Our services

  • Assistance in experimental design
  • Sample preparation starting from gateway® entry-clone
(in combination with gateway shuttle service )
  • Viral packaging of expression constructs
  • Transduction of target cell lines
  • Virus clearance
  • Safety handling and documentation (GTAS)

You supply

  • Sequence verified lenti- or retroviral expression constructs (entry clones in combination shuttle service) in midipräp quality (10µg/ transduction)
  • Cell line to be transduced together with a bottle of complete medium.

Generation of viral particles

For biological safety all viral particles produced in the Virus Transduction Service are replication incompetent to prevent the formation of an infectious wildtype virus. For packaging of the expression constructs we use a 2nd-generation system consisting of 3 plasmids. The functional viral components are subdivided onto different plasmids:

  • Lenti-or retroviral transfer plasmid
  • Packaging plasmid (e.g Gag, Pol, Rev, Tat)
  • Envelope plasmid (e.g pVSV-G)

The Virus Transduction Service currently does not intend to produce large amounts of high titer virus stocks or store larger amounts of viral supernatant. Therefore we have to dispose the viral particles 4 weeks after delivery of the transduced cell line.
