Total Internal Reflection Fluorescence Microscopy (TIRF)

Total Internal Reflection Fluorescence Microscopy is based on the evanescent electromagnetic field that is generated upon total reflection of the excitation light at the interface between coverslip and medium. TIRF is a powerful optical tool for the selective visualisation of processes and structures that occur at this interface. Superior signal-to-noise ratios and excellent z-resolution are the characteristics of TIRF which enable the visualisation of even small structures. We use TIRF for molecular studies in cell free preparations on the single-molecule level.
