Proteins play a crucial role in all life processes. They serve as construction materials, transmit messages from cell to cell, and are responsible for regulating metabolism. Proteins are long chains of amino acids. In cancer research, purified proteins are used for a wide variety of applications, including assay development, molecular biology applications, raise of antibodies, binding studies and more. Therefore, protein purification is a first step of almost all in vitro protein studies. High quality of purified protein could be critical in respect to their activity and folding. Proteins can be isolated from their native environment or from recombinant sources like E. coli, yeast, baculo virus or mammalian cell culture.
Although there is no central facility for protein expression or culture fermentation at DKFZ, a unique access to protein purification expertise is offered through cooperation with Prof. Klaus-Jürgen Schleifer, Computational Chemistry and Biology, BASF, Ludwigshafen.
The scientific service group at BASF has outstanding experience and expertise in the purification and analysis of proteins and Edman Sequencing. Standard processes at a scale of 25 ml to 1 l of lysate, include:
- Antibody purification
- His-tag purification
- GST-tag purification
Beside these routine techniques, the offer includes the establishment of novel purification schemes upon request. All state-of-the-art technologies are up and running, and can be combined depending on individual requirements. Column sizes range from 10 ml to 300 ml.

Input from you
As starting material you need to provide protein lysates. Please ensure that proteolytic degradation is inhibited. You may contact the service personal beforehand to discuss purification needs, and to arrange for save transfer of material.
What we do
We perform incoming QC for concentration and size distribution. Upon acceptation of samples we perform the purification scheme agreed. Thi susually includes:

- Buffer exchange if necessary/wanted
- Sterile filtration
- OD measurement
