MS Service

In the characterization of molecular structure, knowledge of the molecular weight is of prime importance. We provide mass spectrometry (MS) service for the molecular weight determination of "small" molecules: e.g., synthetic compounds, natural products, metabolites, carbohydrates, peptides, nucleotides, drugs, metal chelators, etc. In general, we use the techniques of Electrospray ionization (ESI) since this is the most gentle of the soft ionization methods currently available. As a result, the molecular weight determination of intact molecules can be performed even on very labile compounds such as peroxides. For each sample, we record both positive-ion and a negative-ion ESI-MS data. For the majority of compounds, protonated molecular ions [M+H]+ are detected in the positive-ion mode and deprotonated ions [M-H]- are detected in the negative-ion mode. These complementary data sets allow unambiguous recognition of the molecular ion in the presence of ion clusters involving Na+, K+, Cl-, etc.

MS Research & Development

Research is focused on method improvement for analysis of protein phosphorylation by nonradioactive methods. For this purpose, we have available in-gel digestion methods with various proteases, nanoESI-MS/MS on a triple-quadrupole system and on a hybrid Q-TOF instrument, capillaryLC-ESI-MS on both instruments, enzymatic and chemical dephosphorylation micromethods, IMAC microenrichment for phosphopeptides, and capillaryLC-element mass spectrometry with phosphorus detection. We are working towards an optimally sensitive combination of these methods to provide complete phosphopeptide coverage and identification of phosphorylation sites for gel-isolated proteins.
