Part I – Theory

· Genetic alteration: risk and chance
· Usage of transgenic mice
· Injection of DNA into pronucleus of zygotes
· Embryonic stem cells and homologous recombination
· Cre/loxP system
· Injection of embryonic stem cells into blastocysts
· Alternative methods
· Cryopreservation of embryos and sperm from mice
· Vasectomy of male mice
Part II – Practice (demonstration)

· Introduction into isolator housing system
· Collection of mouse embryos (zygotes and morulae)
· Injection of embryonic stem cells into blastocysts and morulae
· Injection of DNA into pronucleus and cytosol of zygotes
· Transfer of embryos into the uterus
Participation of part I is necessary for visiting the practical part.
