Part I – Theory

· Transgenic animal models - for what reason?
· General strategy for generation of transgenic mice
· DNA microinjection into pronucleus of zygotes
· Embryonic stem cells and homologous recombination
· Injection of ES cells into blastocysts
· Comparison of DNA microinjection versus ES cell injection
· Vasectomy of male mice
· Cryopresservation of mouse embryos and sperm
. Cre/loxP and Flp/FRT system
· Alternative methods for generation of transgenic mice

Part II – Practice

· Introduction into animal (mice) housing
· Collection of mouse embryos (zygotes and blastocysts)
· Injection of DNA into pronucleus of zygotes
· Injection of embryonic stem cells into blastocysts
· Transfer of embryos into the oviduct and uterus
· Vasectomy of male mice

Participation of part I is necessary for visiting the practical part.
